Monash Gene Targeting Facility

The Monash Gene Targeting Facility is a core facility providing gene targeting services in mice to the academic researcher community. The facility is located at the Monash Institute of Medical Research and includes capabilities in molecular biology, tissue culture, embryology and animal husbandry.  The scientists in the facility have several years experience in producing genetically modified mice and the operation is overseen by an advisory committee with many years of experience and expertise in this area. The facility has capabilities in:

Molecular biology:
Design of targeting and screening strategies
Design and construction of gene targeting vectors
Screening of embryonic stem cell clones for targeting events
Genotyping to identify animals that are heterozygous for the targeted allele

Tissue culture:
Electroporation of the targeting vector into embryonic stem cells
Selection for targeted embryonic stem cell clones
In vitro recombinase treatment (cre or flp) to remove the selection cassette if required

Embryology/animal husbandry:
Microinjection of targeted embryonic stem cell clones into blastocysts and transfer to recipients
Generation of chimeras and breeding to obtain germ line transmission of the targeted allele
